5F-ADB (alternatief gespeld als 5F-MDMB-PINACA) is een synthetische cannabino\u00efde afgeleid van indazol die is gebruikt als een actief ingredi\u00ebnt in synthetische cannabisproducten en online wordt verkocht als een designerdrug.<\/p>\n

5F -ADB is een sterke agonist van de CB1-receptor, hoewel de selectiviteit voor deze receptor onbekend is. 5F-ADB werd voor het eerst gevonden in november 2014 met behulp van postmortale monsters die waren verkregen van een persoon die stierf na het nemen van een product dat dit medicijn bevat. Daaropvolgende tests onthulden dat 5F-ADB werd gedetecteerd bij in totaal tien mensen die tussen september en december 2014 in Japan stierven door onverklaarbare overdoses. Om de neuronale mechanismen achter 5F-toxiciteit op te helderen, bestudeerden we de effecten ervan op dopaminerge en serotonerge circuits in de middenhersenen, die een verscheidenheid aan fundamentele hersenfuncties reguleren, inclusief die geassocieerd met beloningsgerelateerd gedrag. Ex vivo elektrofysiologische technieken werden gebruikt om veranderingen in de spontane activeringsactiviteit van dopaminerge en serotonerge neuronen geproduceerd door 5F-ADB vast te leggen.<\/p>\n

5F-ADB (1 M) verhoogde de spontane activeringssnelheid van dopaminerge neuronen dramatisch, maar slaagde er niet in ze te activeren in aanwezigheid van de CB1-antagonist AM251 (1 M). Dezelfde concentratie van 5F-ADB had daarentegen geen effect op de serotonerge neuronactiviteit.
